Figure 2 Engraftment of bone marrow-derived cells in the gastric mucosa: effects of time and infection. C57BL/6 mice were myloablated and underwent transplantation with marrow from wild-type C57BL/6 or ROSA-26 mice carrying the β-galactosidase gene. After recovery, mice were infected with Helicobacter felis or vehicle control. Engrafting cells were detected using x-gal reagent, which produces a blue cytoplasm in cells expressing the β-galactosidase enzyme. (A) C57BL/6 mice that underwent transplantation with C57BL/6 marrow, infected with H felis for 30 weeks, do not show reactivity with x-gal reagent (blue). Larger area from the same mouse shown in F. (B) C57BL/6 mice that underwent transplantation with ROSA-26 marrow and sham infected or, in C, infected with H felis for 4 weeks demonstrate marrow-derived (blue) inflammatory cells. We did not demonstrate any gastric epithelial glands, which were bone marrow derived. (D) ROSA transplantation mice infected for 20 weeks had few marrow-derived glands (blue, arrow). (E) By 12–18 months of chronic infection, large numbers of glands were completely replaced by cells that were marrow derived (blue). (G) Low-power view of the area.
