Figure 1 Identification of USP7 as a PCNA-Deubiquitinating Protein In Vitro (A) Scheme of purification of the deubiquitinating activity from HeLa cell extracts. (B and C) Detection of the deubiquitinating activity in column chromatography fractions of MonoQ (PC1.6/5) (B) or Superdex 200 (C). The reactions were subjected to western blotting with an anti-PCNA antibody. Minus sign (−) indicates buffer; In, input fraction before column chromatography. PCNA-Ub, mono-ubiquitinated PCNA. (D) A portion of the Superdex 200 fractions was subjected to SDS-PAGE and stained with Coomassie brilliant blue. The arrow indicates the 140-kDa polypeptide co-eluted with the activity. M, protein ladder marker. (E) USP7 has an intrinsic deubiquitinating activity toward mono-ubiquitinated PCNA. Mono-ubiquitinated PCNA was subjected to the standard deubiquitinating assays with the indicated concentrations of His6-USP7. Figure 2 Contribution of USP7 to the Suppression of UV- and H2O2-Induced PCNA Mono-ubiquitination in Human Cells (A) Suppression of USP7 or USP1 increases the level of UV-induced PCNA mono-ubiquitination with different kinetics. Forty-eight hours after transfection with the indicated siRNA, WI38VA13 cells were treated with 20 J/m2 UVC and harvested at the indicated time points. Whole-cell extracts were analyzed by western blotting using antibodies against the indicated proteins. si, small interfering; Ctrl, control; PCNA-Ub, mono-ubiquitinated PCNA; exp, exposure. (B) Suppression of USP7, rather than USP1, increases H2O2-induced PCNA mono-ubiquitination. The siRNA-transfected cells were treated with 0.5 mM H2O2 and analyzed at the indicated time points. (C) The PCNA K164R mutant does not undergo PCNA mono-ubiquitination induced by USP7 suppression after H2O2 treatment. WI38VA13 cells stably expressing HA-PCNA WT or the HA-PCNA K164R mutant were transfected with the indicated siRNAs. The cells were treated with 0.5 mM H2O2 for 20 min, and then whole-cell extracts were analyzed by western blotting using anti-HA, anti-USP7, and anti-Lamin B antibodies. IB, immunoblot. (D) Suppression of RAD18 abolishes the increase in PCNA mono-ubiquitination induced by USP7 suppression after H2O2 treatment. The siRNA-transfected cells were treated with 0.5 mM H2O2 for 20 min, and whole-cell extracts were analyzed by western blotting. See also Figure S1. Figure 3 The Contribution of USP7 to the Suppression of PCNA Mono-ubiquitination Is Cell Cycle Independent, in Contrast to That of USP1 (A) Scheme of the experiment. h, hours. (B) Cell-cycle profiles. Before H2O2 treatment, cells were analyzed by flow cytometry. si, small interfering; Ctrl, control. (C) Suppression of USP1, but not USP7, increases the level of UV-induced PCNA mono-ubiquitination during S-phase. Thirty minutes after UV irradiation or mock treatment, whole-cell extracts were analyzed by western blotting using antibodies against the indicated proteins. PCNA-Ub, mono-ubiquitinated PCNA; exp, exposure. (D) Suppression of USP7 or USP1 increases PCNA mono-ubiquitination with different patterns during the cell cycle. Whole-cell extracts from mock-treated or H2O2-treated (20 min) cells were analyzed by western blotting. Figure 4 Exclusive Contributions of USP7 and USP1 to Cellular Responses to H2O2 Treatment and UV Irradiation (A and B) Cellular sensitivities. WI38VA13 cells transfected with the indicated siRNAs were treated with indicated doses of UVC (A) or H2O2 (B). Data represent the means ± SD of at least three independent experiments. si, small interfering; Ctrl, control. (C and D) Mutagenesis. HAT (hypoxanthine, aminopterin, and thymidine) pre-selected WI38VA13 cells transfected with the indicated siRNAs were treated with 8 J/m2 UVC (C) or 250 μM H2O2 for 1 hr (D). Data represent the means ± SD of at least three independent experiments. See also Figure S2, Table S1, and Table S2.
