Assessing the effect of TNF-α and TRAIL on A2-PPI15–24 CD8 T-cell cytotoxicity. Assessing the effect of TNF-α and TRAIL on A2-PPI15–24 CD8 T-cell cytotoxicity. A: A humanized anti–TNF-α antibody was used in cytotoxicity assays to block TNF-α produced by A2-PPI15–24 CD8 T-cell clones in coculture with K562-A2 target cells presenting cognate PPI15–24 peptide. No inhibition of target cell lysis by the A2-PPI15–24 CD8 T-cell clone was detected with blockade of soluble TNF-α. B: Using blocking anti-TRAIL antibody in cytotoxicity assays does not inhibit A2-PPI15–24 CD8 T-cell cytotoxicity. Bars represent means from triplicate assay wells and error bars SEM. Cytotoxicity assays were performed at an effector/target ratio of 25:1 for 4 h. CTL, cytotoxic T lymphocyte. Robin R. Knight et al. Diabetes 2013;62:205-213 ©2013 by American Diabetes Association
