Under feeder cell-free, iPSCs were cultured at a density of about 30 colonies per 100-mm dish. When the individual colony reached approximately 1 mm in diameter (day 0), BMP4 (80 ng/mL) was added to the mTeSR1 culture medium for 4 days (days 0-4). In turn, the culture medium was replaced with StemPro-34 serum-free medium, supplemented with 1% nonessential amino acids, 2 mM L-glutamine, 100 U/mL penicillin, 100 μg/mL streptomycin (referred to as MC basal medium), and a cytokine cocktail of 80 ng/mL VEGF, 25 ng/mL bFGF and 100 ng/mL SCF for 2 days (days 4-6). The protocol for hematopoietic progenitor expansion and initial myeloid differentiation was modified from Yanagimachi et al. (32). Cells were cultured in MC basal medium supplemented with 100 ng/mL SCF, 10 ng/mL IL-3, 10 ng/mL TPO, 100 ng/mL IL-6, and 10 ng/mL FL. The medium was replaced every 3-4 days, and nonadherent cells were collected after 3 weeks (from day 27 onward).
