Arrows show the positions and orientations of IS6110 insertions. Left- and right-oriented arrows on the maps indicate IS6110 insertions, confirmed by insertion site mapping at the Public Health Research Institute Tuberculosis Center (unpublished data) and the positions of IS6110 according to Beggs et al. (18). The coordinates of the IS6110 insertions in all three strains correspond to the H37Rv annotated sequence. IS6110 mapping indicates that despite the insertion-preferential loci (“hot spots”), the precise positions (flanking sequences of the IS element) and orientations of the inserted sequences are not repeated in the three strains analyzed; i.e., none of the flanking regions of the four IS6110 copies in CDC1551 correspond to the 17 insertion sites in H37Rv or to the 23 positions (resulting in 21 hybridization bands) of the IS element in strain 210. The structures of the DR loci within these three strains are shown; black dots indicate spacers in the DR locus of corresponding strains, and triangles indicate deleted spacers.
