BHLHB2 is part of a protein–DNA complex associated with the proximal E-box element of Bdnf promoter 4. Autoradiograph showing a specific band indicative of a DNA–protein binding complex in NIH3T3 cell nuclear extracts to a 33P-labeled double-stranded DNA probe based on the 22 bp rat Bdnf promoter 4 IS region (−110 to −87). Addition of unlabeled competitor DNA caused the binding activity to disappear in a concentration-dependent manner. Other consensus E-box containing competitor DNAs were also effective in eliminating the specific band. When a polyclonal BHLHB2 peptide antibody was incubated with the DNA–protein complex, the migration of the specific band was further retarded (shown by the arrow labeled “supershift”).
