Figure 1 Tm-22 Expression Level Determines TMV Resistance Type in Transgenic Plants. AResponse of Tm-22-transformed N. benthamiana plants to TMV–GFP infection. Symptoms of local and systemic leaves in Tm-22 transgenic plants (TM#1, 34, and 74) and wild-type plants (WT) after TMV–GFP infection. Green fluorescence foci marked the TMV-expressed GFP. Left panels show the local leaves at 5 dpi. Right panels show the systemic leaves at 9 dpi. All leaves were photographed under white light and UV light. BRT–PCR was performed using TMV–GFP CP-specific primers to test the presence of TMV–GFP in local (5 dpi) and systemic leaves (9 dpi) of four plants (WT, TM#1, 34, and 74). CTm-22 mRNA levels in transgenic ER, HR, and SHR plants (TM#1, 5, 13, 20, 34, and 74) were analyzed by real-time RT–PCR using Tm-22-specific primer and normalized relative to the value of NbActin mRNA. ** P < 0.05; *** P < 0.01. Systemic leaves of TM#1 (ER) and TM#34 (HR) Tm-22-silenced plants and respective TRV vector control plants were photographed at 20 dpi under white light and UV light. White arrows indicate SHR. ERT–PCR was performed using TMV–GFP CP-specific primers to confirm the presence of TMV–GFP in the systemic leaves of Tm-22-silenced ER and HR plants 20 dpi. TRV alone-infected control plants were used as negative control. FTm-22 mRNA levels in Tm-22-silenced ER and HR plants were analyzed by real-time RT–PCR using Tm-22-specific primers and normalized relative to the value of NbActin mRNA.
