MCF-10A cells were treated for 6 days with E2 (1 μmol/L) in the presence and absence of SERMs (1 μmol/L). Cell media were analyzed for 2-MeOE1 and 4-MeOE1 formation using LC/MS-MS and an internal standard. “% MeOE1” was normalized to 100% representing cells treated with E2 alone. Attenuation of 4-MeOE1 formation with DMA or Ral cotreatment was significant; FDMA had no effect. Each data point represents an average of 3 independent experiments ± SD. *, P < 0.05.
