Presentation transcript:

Immunization with F/T cells elicits CTLs with moderate and transient effector function. Immunization with F/T cells elicits CTLs with moderate and transient effector function. (A) Experimental design for determining the development of CTL effector function following immunization with IR or F/T 4T1-OVA cells. Groups of C57BL/6 mice were adoptively transferred with OT-I T cells (2.5×105/mouse) 1 d prior to immunization with IR or F/T 4T1-OVA cells. At the indicated time points, all mice received a 1:1 mixture of target cells (SIINFEKL-pulsed CFSEhigh:unpulsed CFSElow splenocytes) as described in Materials and Methods. Mice were harvested 20 h post-transfer and the DLNs were analyzed for specific killing of target cells. (B) Kinetics of effector function acquisition by CTLs following immunization with IR or F/T 4T1-OVA cells. Data are shown as the mean percent specific killing compared to mice immunized with PBS. Results from 3 to 4 independent experiments were averaged per time point and error bars represent SD between samples. (C) CTL activity in vivo in DLNs and spleen, 9 d following immunization with IR or F/T 4T1-OVA cells. Matthew R. Buckwalter, and Pramod K.
