Microscope Use What NOT to do with a microscope: Never use the coarse adjustment knob when using the high-power objective Never put drops on a slide that is resting on the stage. Never carry the microscope with only one hand. Never carry by the eyepiece/ocular. Calculating Magnification: - Eyepiece is 10x - Objectives are 4x, 10x, 40x

Steps to MAKE a Wet-Mount Slide Obtain materials: Water Dropper/Pipette Specimen/Sample Slide Coverslip Clean slide with a Kimwipe Put one drop of water on slide (only if specimen is not already in water) Put specimen in drop of water

Steps to MAKE a Wet-Mount Slide 5. Put coverslip on using the following steps: Touch coverslip to slide at a 45 angle. Drag to drop of water Slowly lower over specimen 6. Clean off any excess water

Steps to STAIN a Wet-Mount Slide If slide is already on stage of microscope, carefully remove it Put one drop of stain on the slide on one edge of the coverslip. Touch the corner of a paper towel to the opposite edge of coverslip Clean up any excess water/stain
