Gram staining was originally developed in 1844 by Christian Gram Because most bacteria have one of these two types of cell walls, we can use this difference as a feature that can be identified using the Gram stain. The Gram stain is a differential stain that uses two dyes to differentiate between the two basic bacterial cell wall types
This stain for differentiate between G +ve bacteria (which accept staining) and G -ve bacteria (which not accept staining).
- Then pour Crystal Violet stain on the slide and leave it for1 minute. - Wash gently with water. - Then pour Iodine stain on the slide and leave it for 1 minute. -Wash gently with water. -Wash with Alcohol to decolorize the slide. -Then pour Safranin stain on the slide and leave it for 1 minute. -Let the smear dry at room temperature or blot with the bibulous paper.
