Bacterial identification procedure Part 2 : Reading of API system

Materials avalaible API system after incubation Computer with bacterial identification software API system after incubation Reagents API results card

1. Gallery validation

First, read the GLU test. A yellow color show a good incubation

2. Add reagents

Four complementary tests must be realized. TDA test : add 1 drop of FeCl³ reagent. IND test : add 1 drop of Kovacs reagent. 6

VP test : add 1 drop of a-naphtol reagent and 1 drop of KOH reagent. NR test : in the GLU test cup, add 1 drop of a-naphtylamin reagent and 1 drop of sulfanilic acid reagent.

3. Results

3.1. For each test observe the colors 3.2. With the API datasheet, deduce if the test is positive or negative.

3.3. Complete test after test, the API results card. …… + - + - + -

4. Results interpretation

4.1. On the computer, open the bacterial identification software. 4.2. Report your results + and – on the software cells. Press « enter » on the keyboard to move cell from cell. 12

4.3. At the end of the procedure, write the name of the bacterial strain proposed by the software. 13

5. Final expression of your results What the name of your bacteria ?
