HYPOTHESIS -CANCER CURE LIES IN GRAVIOLA TREE (SOUR SUP) -ANNONACIN FROM GRAVIOLA TREE CAN INHIBIT MULTI-DRUG RESISTANT CANCEROUS CELLS IN HUMANS.

SPECIFIC AIMS - Using annonacin from graviola tree to eliminate multi-drug resistant tumor/cancerous cells - Fighting against multi-drug resistant cancerous cells -To extract pure sample of annonacin from seeds of graviola tree (sour sop) - Ensuring cancer free environment.

Annonacin is present in the leaves, fruits, stems and seeds of graviola trees. Make sour- sop juice your best friend!!!

INNOVATION -Most of the cancerous cells are becoming more resistance to cancer drugs. -Increase in the number of cancer patients annually. -Elimination of Multi-drug resistant cancerous cells. -To make our environment cancer free.

SIGNIFICANCE -Annonacin has the ability to inhibit multi-drug resistance cancerous cells. - It can eliminate cancer multi-drug resistance cells more faster than other cancer drugs.

Annonacin - class of compounds known as acetogenins Annonacin - class of compounds known as acetogenins. - characterized by linear 35carbon chains containing oxygenated functional groups including hydroxyls, ester, epoxides. Formula: C35H64O7

Extraction and purification of annonacin -dried and powdered seed (671 g) of A. muricata were extracted with methanol at room temperature (20C) to give a brown extract (60 g). - concentrated methanol extract was partitioned between dichloromethane and water to yield 45 g of dichloromethane extract, that reacted positively with the Kedde reagent (10% 3,5-dinitho- benzoic acid in ethanol, followed by 2 N KOH in methanol), indicating the presence of unsaturated lactones. -This extract was subjected to silica gel column chromatography (silica gel 60 Merck, 70–230 mesh) and was eluted with n-hexane containing gradually increasing amounts of ethyl acetate.

CONT’D -The fractions collected were analysed by TLC (silica gel Merck 60 F254), on the basis of which they were grouped into 17 pooled fractions. -The solvent from fraction 15 was removed, and the resulting residue (12.2 g, Kedde positive) was further subjected to column chromatography (silica gel 60 H Merck; mobile phase dichloromethane/ isopropanol 90/10) to yield crude annonacin (840 mg). -The purity of this compound was confirmed by an analytical HPLC procedure using a reversed-phase Waters, Pure annonacin was confirmed.

MECHANISM OF ACTION OF ANNONACIN -Multi-drug resistant cancerous cells form pump -Blocking of larger pumps formed by cancerous cells -These pumps are called P-glycoprotein
