An introduction to a novel filtration system ATF-manufacturing Platform Upstream Unit Operations ATF-cellcultivation Concentrated Perfusion Concentrated Fed-Batch Hybrid Processes

UF filter concentrates protein inside reactor Filtrate pump runs at vv / day or as required by cell growth ATF rate constant at ~ vv / min (or faster) Feeding controlled by level probe Concentrated Fed-Batch (CFB) Has the benefits of Concentrated Perfusion (CP) and Fed-Batch together –high cell concentration (70-200m/ml) achievable in 2 weeks Straight-forward feeding control & removal of unwanted metabolites Concentrated harvest in one identified batch: CHO at 17g/L (and higher) CFB allows retention of some protecting proteins and growth factors so viability may be higher than CP, removing the need for a bleed and simplifying the process control
