YEAST LAB DEAD YEAST W/MINERAL OIL 2 mL of sucrose solution to 50 mL beaker Add 2 mL of DEAD yeast; pipette up and down 2-3x to mix Cover this solution with mineral oil Incubate beaker in warm water bath (38°-42°C) for 5 min Place both beakers into biochamber, insert probe and press collect (data should collect every 15 seconds for 4 minutes) After data collection, restore warm water bath temperature Fan air over bio-chamber to restore gas levels

YEAST LAB DEAD YEAST W/O MINERAL OIL Pipette the yeast solution from under mineral oil to clean 50 mL beaker (better to transfer less yeast than to transfer mineral oil) Incubate beaker in warm water bath (38°-42°C) for 5 min Place both beakers into biochamber, insert probe and press collect When asked, choose ‘store latest run’ Data should collect every 15 seconds for 4 minutes

YEAST LAB LIVE YEAST W/MINERAL OIL 2 mL of sucrose solution to 50 mL beaker Add 2 mL of LIVE yeast; pipette up and down 2-3x to mix *Be careful, pipetting too vigorously will kill the yeast Cover this solution with mineral oil Incubate beaker in warm water bath (38°-42°C) for 5 min Place both beakers into biochamber, insert probe and press collect, click ‘store latest run’ (data should collect every 15 seconds for 4 minutes) After data collection, restore warm water bath temperature Fan air over bio-chamber to restore gas levels
