 To familiarize with the use of oceanographic instruments  To assess the water quality, distribution and diversity of plankton and benthos  To evaluate how gradient of organic pollution affects the distribution and abundance of phytoplankton and zooplankton

3 sites 6 stations on board

Global positioning system (GPS) Exact coordinates of sampling station (before and after sampling) Water depth (3 replicates) Use the echo sounder to measure water depth Water sampling Collect from: Surface (x1) Nutrients Surface (x1),Middle (x1) and Bottom (x1, 1 m above bottom) Salinity, pH Jot down bottle number Put bottle into the ice box

 Zooplankton sampling - Haul the zooplankton net (125 μm mesh size) from 2m above bottom to surface - Rinse plankton net with filtered seawater - Store and preserve sample with 35% formalin ( 5 drops) in white bottle - Write down bottle number

 Phytoplankton sampling - Haul the phytoplankton net (80 μm mesh size) from 4 m deep to surface - Rinse plankton net with filtered seawater - Store and preserve sample with Lugol’s solution ( 5 drops) in dark bottle -Write down bottle number

 Chlorophyll content - Collect surface water with bucket - Filter 200 ml water - Add 5 drops of MgCO 3 (shake before use) - Store the filter paper in a labeled 15 ml centrifugal tube (label Group, Site & Filtered Volume) - Wrap with aluminium foil  icebox - Pass the filtered seawater to Station 2 & 3

 Water clarity - Measure water clarity with Secchi disk (do it 3 times)  Benthic organisms sampling - Use mud grab to collect the bottom sediment - Sieve to search for living organisms - Preserve benthos with ethanol in labeled sample bottles (Group & Site)

 Water temperature and Dissolved Oxygen (DO) Use the multimeter to determine water temperature and DO concentration at 2 m interval when water depth > 10m or1 m interval when water depth < 10m Measure from 1 m above bottom to surface
