Different Kinds of Microscopes Learning how to use the microscope

Learning Objectives  I can use a compound light microscope to draw and describe observations about cells.  I can explain why the microscope was an important invention/technology to advance human understanding of life.  I know that further advances in microscope technology continue to advance human understanding of life/cell's processes.

There are 2 kinds of microscopes: one lensmagnifying Simple: one lens (magnifying glass) Compound: 2 or more lenses

Simple vs. Compound VideoClip

Learning Parts of the Microscope

Parts of the Microscope Revolving Nosepiece 1. Revolving Nosepiece

Parts of the Microscope 2. Lens/Objective

Parts of the Microscope 3. Stage

Parts of the Microscope 4. Disc Diaphragm (controls amount of light) 1 = lowest setting/ 1 = lowest setting/ 5 = highest setting

Parts of the Microscope 5. Light source

Parts of the Microscope 6. Base

Parts of the Microscope 7. Eyepiece

Parts of the Microscope 8. Arm

Parts of the Microscope 9. Stage Clips

Parts of the Microscope 10. Course Focus/ Adjustment

Parts of the Microscope 11. Fine Focus/ Adjustment

Calculating Magnification Multiply lens powers together!

Eyepiece10 XEyepiece: 10 X by itself Low Power:Low Power: 4red 40 X large objects/scanning4X; red color; and a TOTAL magnification of 40 X mainly used for large objects/scanning

Medium Power: 10 X, yellow 100 XMedium Power: 10 X, yellow color; TOTAL magnification of 100 X High Power: 40 X;blue 400 X; mainly used for smaller objects/detailsHigh Power: 40 X; blue color; and a TOTAL magnification of 400 X; mainly used for smaller objects/details

Parts of the Microscope Clip

When you are ready to start: LOW Make sure it is on LOW power! down The stage should be all the way down (you focus by slowly moving it up!) 1-2 to start Check the light setting (should be at 1-2 to start)

Other Things to Consider: Carrying the Microscope: Always use 2 hands!

Other Things to Consider: HIGH Never use the COURSE FOCUS KNOB while on HIGH power.

When putting the microscope away (at the end of the day): LOW It should be on LOW power. WRAPPED The cord should be WRAPPED around the base. DOWN The stage should be all the way DOWN (do this with the course focus knob!) OFF The light switch should be OFF

TROUBLESHOOTING What if you can’t get your object into focus? Check that you have one of the lenses clicked into place IN BETWEENCheck that your disc diaphragm is in place (not IN BETWEEN settings)

TROUBLESHOOTING What if you can’t get your object into focus? LENS PAPERClean off your lenses using LENS PAPER. (Also, dirty slides are hard to focus on; so clean them, too!) GO BACK TO LOW POWER AND START OVER!

Procedure For Using the Microscope Clip

MAGNETICUse a MAGNETIC field to bend beams of light; used to see things at much higher level of detail 1,000,000Can magnify images up to 1,000,000 times!

There are 2 kinds: SURFACESSCANNING ELECTRON MICROSCOPES (SEM): Allows you to see the SURFACES of objects

There are 2 kinds: THROUGHTRANSMISSION ELECTRON MICROSCOPES (TEM): Allow you to see THROUGH an object.

Microscopes by Brainpop 1. Why does the microscope stage have a small hole in it? 2. Why are specimens very thin pieces of material? 3. Will we be using an electron microscope in class? Why not?
