Friday September 18th In Notebook: What experience do you have using a microscope? What are some practical uses of a microscope that you are aware of?
Today’s Learning Targets Agenda: 1.Microscope Notes 2.Microscope Lab DUE: Tuesday Sept. 22nd
What’s my power? To calculate the power of magnification, multiply the power of the ocular lens by the power of the objective. Eyepiece is 10x and objective is 10x what is total magnificaion?
Basic Rules and Tips Carry using arm and base Flat Surface Adjust light using diaphragm Use stage clips Focus with course adjustment on low and medium power only! – High Power use fine adjustment only
How to make a wet-mount slide … 1 – Get a clean slide and coverslip from your teacher. 2 – Place ONE drop of water in the middle of the slide. Don’t use too much or the water will run off the edge and make a mess! 3 – Place the edge of the cover slip on one side of the water drop. 4 - Slowly lower the cover slip on top of the drop. Cover Slip Lower slowly
Monday September 21st In Notebook: How do you calculate total magnification of an object through a microscope?
Today’s Learning Targets Agenda: 1.Microscope Notes 2.Microscope Measuring Lab DUE: ? 3.Friday’s Microscope Lab due TODAY at end of hour 4.REMINDER: Lab report due TOMORROW! (Do not need to print) 5.REMINDER: Quiz TOMORROW and test on THURSDAY!
1. Clear ruler 2. Determine how many millimeters across the field. (1 millimeter is the distance from the middle of one mark mark to the middle of the next mark) 3. Convert to micrometers (1mm=1000μm) Calculate Field of View
Examples: 1. If it’s 1.5 mm across, then the field of view is: 2. If it’s 2.4 mm across, then the field of view is:
DOES THE FIELD OF VIEW SIZE CHANGE WITH DIFFERENT MAGNIFICATIONS? Low power = 4 mm or 4000 μm Medium power = 1.5 mm or 1500 μm High power =.5 mm or 500 μm
Calculate Size of Specimen 1.Figure out how many will fit across the field of view (estimate) 2.Divide field of view length by how many fit across Example: If 7 protozoa fit across my field of view with a length of 1400μm, then what is the size of one protozoa?
Practice Looking through my low power objective I see 4 mm on the ruler. Later I was looking at my hair and I estimate I could fit 10 hairs side by side in my field of view. How wide is my hair?
