Recap… Needed to generate concentration curve to determine protein concentration on chips Spotted pure GST protein onto slides in different [] and measured resulting signal. Remember problems: antibody [] too high…

Moving on… We are now searching for a useful yet unique application for our chip Translational control protein specific 380 proteins will go through my purification process and be spotted

What’s the point? Therefore, instead of measuring ALL yeast proteins, we only study those that have a part in translational control. Benefit? Can easily determine effect of one protein against all others without doing unnecessary work.

Future Aims Have received interest from Dr. Walter Chazin of the Vanderbilt Struct. Bio Dept. for the purification/chip development for entire yeast genome. Will study calcium interactions.

More Future Aims During printing processes, will study many different slide surface chemistries Some will have higher binding affinities than others First lab to use Glutathione? Will work with Dr. Link to produce publication detailing these results.

MALDI detection Still hope to apply chip to MALDI mass spec. Will use translational control chip for this now.
