Isolation of Hydrocarbon-degrading Bacteria from Guimaras Soil Samples Soil samples from various oil-contaminated areas in Guimaras will be obtained and stored in 50 mL conical tubes at room temperature. To isolate possible hydrocarbon- degrading bacteria from each sample, a pinch of soil will be inoculated into 50 mL of liquid minimal medium (for 1 L: 0.5g KH 2 PO 4, 2.0g KNO 3, 1.0g MgSO 4 7H 2 O, g CaCl 2 2H 2 O, 0.002g FeSO 4 7H 2 O) with 50 μL of bunker oil (Petron Corporation, Philippines), and subjected to incubation while shaking at 37ºC at 225 rpm for one week. Once a visible change in turbidity is observed, an applicator stick will be dipped into the suspension culture and streak plated on Luria-Burtani (LB) agar (Pronadisa, Madrid, Spain). One streak plate will be made for each soil sample and incubated overnight at 37°C. Isolated bacterial colonies of different colonial morphologies will be picked and subcultured into separate LB agar slants.
