D Develop set of functional and physiological tests to determine how postflight changes in sensorimotor, cardiovascular, and muscle physiology impact functional performance –Data Collection: Pre-/Post-flight testing only; series of timed muscle performance test Integrated Cardiovascular: Benjamin D. Levine, M.D., Michael W. Bungo, M.D. –Quantify the extent, time course and clinical significance of cardiac atrophy (decrease in the size of the heart muscle) associated with long-duration space flight. This experiment will also identify the mechanisms of this atrophy and the functional consequences for crewmembers who will spend extended periods of time in space. –In-Flight Data Collection: Hardware used: HRF Ultrasound, HRF Actiwatch, HRF Holter Monitor 2, ESA Cardiopres. 48 Hr Ambulatory Monitoring within 1 week of Ultrasound scan. Smith, Ph.D –Measures bone metabolism, oxidative damage, chemistry and hormonal changes, as well as assessments of the nutritional status of the astronauts participating in the study. –In-Flight Data Collection: Blood and 24-Hr urine samples, Food Frequency Questionnaire Pro K: Scott M. Smith, Ph.D. –Evaluation of a dietary countermeasure to lessen bone loss of astronauts. Pro K proposes that a flight diet with a decreased ratio of animal protein to potassium will lead to decreased loss of bone mineral. –In-Flight Data Collection: Controlled/monitored diet with urine and blood samples Investigation Summary 6

Payload Operations Integration Working Group (POIWG) Jan 25-27 th, 2011 Human Research Program (HRP) Blue Bradley Repository: Kathleen McMonigal, M.D. –The National Aeronautics and Space Administration Biological Specimen Repository is a storage bank that is used to maintain biological specimens over extended periods of time and under well-controlled conditions. –In-Flight Data Collection: additional tubes collected in combination with NUTR study blood draw Reaction Self Test: David F. Dinges, Ph.D. –Short task to evaluate reaction times which will monitor the daily effects of fatigue on performance while on board the International Space Station. –In-Flight Data Collection: Testing using reaction feedback software, scheduled on specific days and sleep shifted schedules, and around EVAs. Spinal Elongation: Sudhakar Rajulu, Ph.D. –Provide quantitative data as to the amount of change that occurs in the seated height due to spinal elongation in microgravity. –In-Flight Data Collection: Seated Height and Standing Height logs, photo documentation. Payload Operations Integration Working Group (POIWG) Jan 25-27 th, 2011 Human Research Program (HRP) Blue Bradley Sprint: Lori Ploutz-Snyder, Ph.D. –Evaluate a new integrated resistance and aerobic training program designed to minimize the loss of muscle, bone, and cardiovascular function during ISS missions. –In-Flight Data Collection: Monitoring of training loads on ARED and T2 will be evaluated as assessment of muscle function. Cardiovascular fitness assessment will include a monthly measurement of VO2max. Muscle response in flight will be measured via ultrasound scans of muscle tissue. Subjects will perform the Sprint exercise prescriptions 6 days per week in flight, beginning as early as possible. Treadmill Kinematics: John K. De Witt, Ph.D. –Collect biomechanical data during treadmill exercise on the ISS to determine the appropriate speed and load combinations that may be the best to maintain bone and muscle health. –In-Flight Data Collection: Visual markers will be placed on the crewmember’s body and high definition video will be recorded and downlinked to be analyzed on the ground. Specific exercise prescriptions for each session will be available to the crew, which they will input manually into Treadmill 2. VO2max: Alan D. Moore, Jr., Ph.D. –Documents changes in maximum oxygen uptake for crewmembers onboard the ISS on long-duration missions. –In-Flight Data Collection: Maximal exercise portion added to monthly PFE using ESA Portable PFS Investigation Summary 8

Payload Operations Integration Working Group (POIWG) Jan 25-27 th, 2011 Human Research Program (HRP) Blue Bradley Urine Monitoring System (UMS) UMS is an automated void volume measurement and sample acquisition system UMS operations will include: –Assembly & Validation –Nom Check-out Ops (Subject Req’d) UMS Validation will include volume confirmation and analysis of pre- positioned non-biological samples flown on 19A UMS Nom Check-out Ops will include two subjects’ 2-day urine collection periods UMS Samples will return on ULF7 The ISS Urine Monitoring System (UMS) is an upgrade to the previous Shuttle UMS that is capable of: 1) Collecting and accurately measuring void volume 2) Separating urine from air 3) Allowing acquisition of individual, uncontaminated urine samples. Payload Operations Integration Working Group (POIWG) Jan 25-27 th, 2011 Human Research Program (HRP) Blue Bradley Forward Work –Work with POIC/PLUTO to identify SSC for use inside the WHC for the timeframe of the collections (for the initial “commissioning” – Val/C-O and later for nominal payload collections) –The Emerald brick from SSC20 is specifically allocated for Shuttle middeck flight activities, so if our ops are NOT during docked ops – this is our best bet at getting the emerald brick reqmt filled. –Maintenance flushes of UMS may be required and need to be scheduled at a TBD frequency. Payload Operations Integration Working Group (POIWG) Jan 25-27 th, 2011 Human Research Program (HRP) Blue Bradley Current Implementation Details Support during IP Science use of HRF Hardware will only include a HRF Systems Engineer on console for direction of safe and consistent use of the HRF Hardware; no science or stowage calls should be expected from the HRF Console Direct communication with the crew, when available, is required during the Calibration and Instrument check of VO2max sessions to answer calldowns regarding hardware anomalies. Already a GR&C and proposed ULF-6 Payload Reg. HRP has a contract with Med Trainers to support blood draws. S/G will be tied to private loop prior to activity to allow for crew calldowns regarding the blood draws OCA downlinks: OC to write IFN to action OCA officer to perform downlinks per timeline. Proprietary or sensitive information should not be included in crew notes, IPV state data, or other public avenues of communication OCA Downlink of restricted data will go from OC to OCA to TSC drop box, photos via Bldg 8 Notable 21

Nutrition/Repository/Pro K 24-hr urine collections can be scheduled over the weekend to allow flexibility in timeline management and will start with the first morning void on a given day and end with the void collection after 24-hrs has passed from the initial void. 26S FE3 must have blood OPR, per request. Crew preference for restraint during scan (e.g, CMRS or “bear hug” method) will be communicated to the COL Flight Planning team for awareness of CMRS in Columbus. ICV Ambulatory Monitoring can start on a Friday to minimize interference with finger-cuff data collection. Crew preference for Integrated Immune dry saliva collection reminders – 27S FE6 requested they be hard scheduled. Crew Preferences concerning 24-hr Urine Collection scheduling, constraint reminders, Pill Ingestion scheduling, configuration states will be identified in Crew Increment Status Briefings and relayed to POIC for documentation in stage payload regulations Notable 22

The E26 CDR has received validation testing training; however 25S FE6 is preferred for execution Actiwatch: Download/Initialization/battery changes can be done by any trained crew – Caveat: HRP will plan to utilize the Legacy Actiwatches throughout Inc 25/26 unless the Actiwatch has a non-recoverable malfunction SLAMMD: Set-up and Stow can be done by any trained crew HRF Facility Activities: Most HRP Facility are flexible as to when they need to be scheduled aside from the following: – HRF Supply Kit drawer inserts are launching on ULF5 with re-supplies launching on subsequent flights. Initial crew configuration activity will pull together all required contents and confirm initial configuration with photo documentation. Inventory activity will be performed near the end of Inc 25/26 to allow for planning of future resupply 32
