The two momentary switches -WAIT TAPE will halt the sample wheel rotation and stop recording for tape exchanges, operator will toggle once to the up position prior to tape removal (LED illuminates green), and then up after tape exchange is complete (LED extinguishes, RUNNING LED illuminates) -END RUN will initiate the SAMPLE WHEEL rotation to the zero position. When toggled to the up position, the END RUN LED will illuminate and remain green until the SAMPLE WHEEL has reached the zero position. Depending on the position of the SAMPLE WHEEL when the switch is toggled, the time to zero position may take up to 1 hour. PAYLOAD HARDWARE (cont.)

POWER/DATA Connector -POWER/DATA (J308) will provide the power and data interface between MSG and the EXPERIMENT BOX The Cover Lid -Provides for a level of containment when properly sealed The Camera Mount -Provides the hardware interface for the MSG Video Camera. The camera must be firmly seated within the mount or the micro switches that are in the bottom of the mount will not activate. When securing the camera the crew needs to listen for the click of the micro switches. During activation there is also a check with the POIC for data confirmation that the micro switches are enabled. 18 18 PAYLOAD HARDWARE (cont.) ELECTRONICS BOX The ELECTRONICS BOX is fitted with 3 M6 Captive Screws that are identical to the ones in the PROMISS 2 EXPERIMENT BOX. There are 3 pigtailed harnesses protruding from the ELECTRONICS BOX: POWER CABLE ->MSG (P325) DATA CABLE ->MSG (P311) POWER AND DATA CABLE ->PROMISS 2 EXPERIMENT BOX (P308). The ELECTRONICS BOX is also fitted with one permanent toggle switch (either OPERATION or UPLOAD mode) that enables the Crew to perform a S/W upload, or operate normally the PROMISS 2 EXPERIMENT BOX. The toggle switch is protected by switch guards.

20 20 PAYLOAD HARDWARE (cont.) SOFTWARE UPLOAD cable This cable will only be used in the event that a software upload to the PromISS 2 ELECTRONICS Box is required. The following steps represent the operations required to perform the software upload. –The ground will perform a software upload from the ground to the MLC via the Payload MDM. –The crew will need to connect the SOFTWARE UPLOAD cable to the MLC and the PromISS 2 ELECTRONICS BOX. Place the OPERATIONS/UPLOAD switch in the UPLOAD position. –The ground will send a ground command to transfer the new software load from the MLC to the PromISS 2 ELECTONICS BOX. –The crew will disconnect the SOFTWARE UPLOAD cable, reconnect the MLC and place the OPERATIONS/UPLOAD switch in the OPERATIONS position. The cable was labeled incorrectly and a hand written label was placed on the cable at Baikonur.

23 23 PAYLOAD HARDWARE (cont.) PromISS 3 Balltip L-Wrench The PromISS 3 Balltip L-Wrench is a PromISS specific tool that is used for the fasteners on the Sample Container and Return Container. It is stowed in the upper foam of the PromISS 3 Kit 2.

25 25 PAYLOAD HARDWARE (cont.) SAMPLE CONTAINER (PromISS 3) The SAMPLE CONTAINER consists of two sealed cavities that are fastened together using 12 stainless steel M6 captive screws. Each cavity contains Phase Change Material (PCM) that will stay frozen at 22° C when faced with an external temperature of 30° C. This will allow the sample cells to be kept at a more constant temperature. The six sample cells are placed in the SAMPLE CONTAINER insert. The SAMPLE CONTAINER provides two levels of containment with two separate Viton o-rings. Once opened the two main cavities remain fastened together with two Kevlar cords that link them. The SAMPLE CONTAINER is also fitted with a pressure valve, in order to allow its easy opening into MSG WV. All the captive screws to be handled by the Crew are of Hex6 type.

27 27 PAYLOAD HARDWARE (cont.) SAMPLE CONTAINER insert (PromISS 3) The SAMPLE CONTAINER insert holds the sample cells within the SAMPLE CONTAINER. The insert is removed from the SAMPLE CONTAINER and will be used in the RETURN CONTAINER to hold the samples for storage in the Russian Incubator Aquarius and return on Soyuz. The SAMPLE CONTAINER insert is not labeled.

29 29 PAYLOAD HARDWARE (cont.) SAMPLE CELLS (PromISS 3) The SAMPLE CELLS are contained in the SAMPLE CONTAINER until they are installed on the SAMPLE WHEEL for integration into the EXPERIMENT BOX. There are six samples with six capillaries per sample. Some of the samples are not including capillaries, but their external housing is always identical. There is approximately 8 seconds between capillaries and 10 minutes between experimental cell rotations. Peak power is expected between sample cell rotation.

31 31 PAYLOAD HARDWARE (cont.) SAMPLE WHEEL The SAMPLE WHEEL will contain the six sample cells. The SAMPLE CELLS will slide into place until the ball lock engages. The Crew has to install the 6 sample cells onto the SAMPLE WHEEL. The Crew uses a Locking System that temporarily secures the SAMPLE WHEEL onto the MSG rear wall. The Crew will insert the SAMPLE CELLS one by one onto the SAMPLE WHEEL. Each SAMPLE CELL is numbered to correspond to the numbers on the SAMPLE WHEEL. One complete SAMPLE WHEEL rotation takes approximately 1 hour when PROMISS 2 EXPERIMENT is operating in nominal mode.

33 33 PAYLOAD HARDWARE (cont.) LOCKING SYSTEM The Locking System is used to temporarily secure the SAMPLE WHEEL to the MSG rear wall so that the SAMPLE CELLS can be installed onto the SAMPLE WHEEL.

34 34 The PromISS CLAMP is a mechanical interface between the PromISS EXPERIMENT BOX and the MSG Airlock Lid. It is one of three fasteners that secures the PromISS to the Airlock Lid, the remaining two are fixed to the EXPERIMENT BOX. NOTE: PromISS is installed to the MSG Airlock Lid. 36 36 PAYLOAD HARDWARE (cont.) PromISS 3 TAPES There are twelve digital tapes that will be used to record the PromISS 3 Video. Tape exchanges 1-5 are every 36 hours. Tape exchanges 6- 10 are every 72 hours. Tape exchange 11 is done after 72 hours. Tape 1 is installed during PromISS 2 SETUP, Tape 12 is removed during PromISS 2 STOW.

39 39 PAYLOAD HARDWARE (cont.) PromISS 3 RETURN CONTAINER The PromISS 3 RETURN CONTAINER will be used for sample return on 7S. It has 10 fasteners that secure the lid and a pressure valve. The pressure valve did not get labeled prior to flight, but is operated exactly the same as the SAMPLE CONTAINER pressure valve. The previous slide shows the RETURN CONTIANER flight unit and how it should have been labeled. The PromISS 3 RETURN CONTAINER will be stored in the MSG WV during the PromISS 3 operations. The RETURN CONTAINER has sugar cubes in a sealed cavity in the bottom. These sugar cubes are used to record temperature and acceleration data during the PromISS 3 operations. The insert from the SAMPLE CONTAINER will be placed in the RETURN CONTAINER during stow. The samples will be stored in the RETURN CONTAINER in the Russian incubator Aquarius until they are brought back on 7S.

Once removed from the SAMPLE CONTAINER/RETURN CONTAINER the MSG WV is sealed and air circulation is on (~3.5mbar), so two levels of containment are still maintained 2. During Stow two levels of containment are maintained. The cells on the sample wheel act as a level of containment and the PromISS 2 EXPERIMENT BOX prior to removal from the EXPERIMENT BOX. When the sample wheel is removed from the EXPERIMENT BOX the cells on the sample wheel and the Air Circulation supply two levels of containment. The Air Circulation is not turned off until the sample cells have been secured in the SAMPLE CONTAINER/RETURN CONTIANER and two levels of containment are in place with the cells and the SAMPLE CONTAINER/RETURN CONTAINER. 3. In the event of a water cooling loop failure, the Electronics Box may exceed touch temperature limit values. If water cooling loop failure is detected the MSG Facility will automatically power down the payloads and the operator must wait a minimum of 10 minutes before entering the WV. 4. The Electronics Box provides a High Voltage source to the Experiment Box, however the connector (J308) is inaccessible at all times after PromISS Activation, precluding a potential shock hazard to the operator. 57 57 PAYLOAD DESCRIPTION Science Objective The monitoring of protein crystal growth in a three-dimensional environment to produce a detailed analysis and quantitative interpretation of the relationship between the quality of the crystals and the environment in which they were produced. The techniques allowing three-dimensional refocusing are mandatory to improve the accuracy of the concentration gradients. Two aspects considered for PromISS 3 are: -The convection motions around the crystal created by the gravity and the composition gradient have a direct impact on the crystal quality and on the concentration of impurities. There is a need to understand accurately these phenomena on the basis of three-dimensional experimental data. -The use of digital holographic monitoring is used to diagnose the locations, the shapes and the sizes of the growing crystals.
