Reviewing Molecular Biology AP Biology Chapters 16,17,18 & 20

 Identify 3 genes that could be on in that cell type and 3 genes that could be off and be able to explain why this might be true!  Describe the different levels of gene regulation and why gene regulation may be controlled at more than one level.  Identify and diagram on your paper 4 ways to activate gene expression (turn genes on) and 4 ways to inhibit gene expression (turn genes off) With the cell type you are assigned:

 Myoblast  Bone Marrow Cell  Metastatic Cancer Cell Examples

 What type of organism was the enzyme or process used in?  How and why is the enzyme used in biotechnology?  What can we apply this biotechnology to study? With the biotechnology you are assigned:

 RNAi  Genetically modifying a bacteria  PCR  Cutting up a gene to run a gel Borrowed Biotech

 How do we construct a plasmid from a restriction map? Plasmid cloning AP Biology 2007

Plasmid Map

 Connect the gene to a fluorescent protein like GFP!  Blue White Screen Ways to identify that a gene has been inserted into a plasmid:

Blue White Screen

 What are some challenges to cloning?  From Embryos  From Adult cells  With cloning for therapeutic purposes  With cloning for agricultural purposes  http://www.pbs.org/wgbh/harvest/etc/video.html http://www.pbs.org/wgbh/harvest/etc/video.html Challenges to cloning eukaryotic organisms (GMO’s):

 Human Genome Project  Basic Science:  How do you sequence a small of DNA?  http://www.pbs.org/wgbh/nova/body/sequence-DNA-for- yourself.html http://www.pbs.org/wgbh/nova/body/sequence-DNA-for- yourself.html  How did we get to the entire genome sequence?  Public Genome Project: from NOVA  http://www.pbs.org/wgbh/nova/body/public-genomes.html http://www.pbs.org/wgbh/nova/body/public-genomes.html  Is personalized, genetic medicine in our future?  Personal Genome Project: from NOVA  http://www.pbs.org/wgbh/nova/body/church-genome-vid.html http://www.pbs.org/wgbh/nova/body/church-genome-vid.html How do we know all this information about DNA?

What types of things would lend well to multiple choice? What types of things could fit in best in the experimental/problem solving section? What types of questions would lend well to short answer? What types of questions would be best suited for open response? What types of questions could make up a test on DNA and Gene Regulation? Test 3/13!!
