Event Related Potentials in Sz and across development Abnormal auditory mismatch negativity (MMN) is a core feature of schizophrenia (Sz), and has been associated with subtle disorganization in thought and language. Prior studies suggest a normal developmental trajectory of increase in auditory MMN amplitudes, specifically for pitch or frequency, from childhood adolescence adulthood (Bishop et al., 2011). For visual P1, in response to faces and shapes, there is a normative decrease in amplitude over development, from childhood adolescence adulthood (MacNamara et al., 2015). To gain insight into the potential time course over which MMN deficits develop in Sz, we obtained MMN to unattended duration, pitch/frequency and intensity deviant stimuli (paradigm from Friedman TJ et al., 2012) in both an adult Sz and community-ascertained (ages 6-25) NKI-Rockland developmental cohort (Nooner et al., 2012). Visual P1 to attended low-spatial frequency contrast gratings were also obtained. For all MMN paradigms Sz patients had reduced amplitude at or below that seen in children. Visual P1 in Sz patients was reduced as compared to all stages of development. Consistent with prior studies (Turetsky et al., 2009), duration MMN was associated with clinical severity of thought disorder. Duration and intensity MMN increase in amplitude from childhood through adolescence We replicated prior findings of decreasing amplitude across development for VISUAL P1 (MacNamara et al., 2015). We DID NOT replicate the findings of Bishop et al., 2011 of increasing amplitude for FREQUENCY MMN across development. We used a higher frequency deviant, whereas they used a lower frequency deviant. In Sz, there was an association of PANSS P2 “conceptual disorganization” with duration MMN, consistent with Turetsky et al., 2009. Subtle thought disorder, like duration MMN, is a known early core feature of Sz. Future directions: time-frequency analysis of auditory MMN and visual P1 over development Duration MMN Developmental cohort: N=43, ages 6 to 25 (mean(SD) =17.4(5.5)), community sample (NKI-Rockland Sample: Nooner et al., 2012). They were divided into 4 groups following Bishop et al., 2011: 1)Ages 6-12 (N = 9); 2) Ages 13-16 (N = 11); 3) Ages 17-21 (N = 11); 4) Ages 22- 26(N = 12) Schizophrenia patients had mean (SD) age of 41(11.5) years. Auditory (unattended) Standard (70%): harmonic tones of 500, 1000 and 1500 Hz superimposed; 100 msec, 85 dB, 5 ms rise/fall Duration deviant (10%): 150 ms Pitch/frequency deviant (10%): 10% lower in frequency Intensity deviant (10%): 10 db lower in intensity Visual (attended) Target stimulus (11%): pre-designated animal schematic (mouse press) Distractor (11%): animal that resembled target 100% contrast horizontal gratings (39% sequential probability each) for high (5 Hz) and low (1 Hz) spatial frequency (HSF, LSF). NB: viewing distance of 114 cm, the stimulus field subtended 6.1 · 4.6 degrees of visual angle; stimuli presented centrally against a 50% gray background isoluminant with mean luminance of sinewave gratings, with light/dark contrast of 80%. SOA were 875 ms ± 25 ms. Auditory/visual stimuli presented concurrently; 250 s blocks, 8 blocks/participant. Continuous EEG data with digital timing tags acquired using a 64-channel ANT with standard reference and ground procedures. Sample rate of 512 Hz. Eyeblinks removed using ICA component analysis in EEGLAB (<10%). Using BESA, data were epoched (-500 to 1000 msec) with baseline (-500 to 0) correction, then high-pass filtered (0.30 Hz, 6 db/oct, forward) and artifacts rejected at +-120 μV, resulting in ~68% of 250 trials retained. Occasionally, channels were interpolated (<10% ). Epochs were averaged off-line for each stimulus type for each participant. MMN was defined as deviant minus standard difference waves. For visual ERP, only P1 for LSF stimuli were evaluated. Latency ranges for duration, intensity, and frequency deviants were 130-230 m, 130–200 ms, and 90– 190 ms post-stimulus onset. Amplitudes measured at frontocentral electrodes relative to average mastoids. Visual P1 to LSF stimuli was obtained in the range of 90-125 ms. Electrodes for each component were chosen based on visual inspection of data and published distributions of auditory and visual ERPs (Friedman et al., 2012; Bishop et al., 2011; MacNamara et al., 2015).
