Today in Room 326/311… Date:2/12/16Topic:Enzymes Day 2Page # ___ HW: Have a relaxing and safe vacation! CONTENT OBJ: SW investigate the effects of temperature, and substrate/enzyme concentration on the rate of enzyme activity LANG. OBJ: SW write a lab protocol of their own creation and discuss in pairs ANNOUNCEMENTS: Website updated if you need… AGENDA: Biomolecules Quiz (30 min) Catalase Demo/Info (10 min) Write Lab Protocol (10 min) Online Lab: Notes/Graphing (30 min)
4
Enzyme Demo Hydrogen Peroxide Yeast contains catalase
5
Demo Yeast & Hydrogen Peroxide Hypothesis: What will happen when we combine them if the chemical equation is this:
6
Factors Affecting Reaction? What factors could cause the rate of the reaction to change? (Increase or decrease) Think about it: person eating a hamburger What would cause him to eat (break down the meat) at a faster rate or slower rate? Now what about the enzyme catalase?
7
Possible Experiments Test the affect of… -Temperature -Substrate concentration (amount of hydrogen peroxide) -Enzyme concentration (amount of yeast) …on the rate of enzyme activity
8
Lab Overview Testing the Factors that Affect Enzyme Function in Catalase
9
Materials List Using the materials available and keeping the possible variables in mind, design an experiment! Group 1: Effect of Temperature on Enzyme Activity Group 2: Effect of Substrate Concentration on Enzyme Activity Group 3: Effect of Enzyme Concentration on Enzyme Activity
10
Lab Protocol: In Journal Each group is assigned a variable Plan to use 3 test tubes (one as a control) Design a lab protocol to test your variable

Online Enzyme Lab Worksheet: Factors Affecting Enzyme Activity (see website for download) Follow this website: http://www.kscience.co.uk/animations/model.swf http://www.kscience.co.uk/animations/model.swf Complete all trials and then draw graphs Complete all trials and then draw graphs
13
Enzyme Limits Our results showed that as enzyme concentration increased, so did the rate of reaction Does that trend go on indefinitely? If we added TONS of enzyme, would the reaction just happen faster and faster and FASTER???
14
Enzyme Concentration We know from our experiment that as enzyme = the reaction rate More enzyme = more chances to collide with substrate But eventually, what happens? Reaction levels off, because not all enzymes can find substrate!
15
Substrate Concentration* What about substrate concentration? Does the rxn rate increase go on indefinitely?? No, eventually the enzyme becomes saturated, meaning all enzymes are tied up with substrate
16
Temperature* What did the results from the experiment show, when testing cold, room temp, and heated liver? Enzymes have an ideal temperature range, because they are proteins When proteins get too hot, they denature (3D structure destroyed = unable to function), and the reaction stops
17
Different Ranges* Different enzymes have different ideal temperature ranges What are the names of ORGANISMS that live in thermal hot springs?? Extremophiles!
18
pH* Just like temperature, enzymes have an ideal pH where they work best (ie the top of the graph) Write on back of sheet!!
