Taking this into account, it makes sense that the induction of pluripotent stem cells (iPS cells) from somatic cells also requires transformation by the two tumor-associated gene products, c-Myc and KLF4 (Figure 3). The Myc protein can elicit various aspects of transformation (Adhikary and Eilers, 2005). However, it also elicits p53-dependent apoptosis in primary fibroblasts. KLF4 might therefore be required to suppress p53 and c-Myc- induced apoptosis

First by viral expression of the transcription factors of Oct4, Sox2, Klf4, and c-Myc The use of genome-integrating viruses could cause insertional mutagenesis and unpredictable genetic dysfunction
5
we attempted to deliver four reprogramming proteins—Oct4,Sox2, Klf4, and c-Myc directly into somatic cells
6
A major hurdle for intracellular delivery of macromolecules such as proteins is their limited ability to cross the cellular membrane
7
Different combinations of transcription factors and small molecules can greatly enhance iPS cells efficiency of reprogramming
9
Here we report that a natural compound, vitamin C (Vc), enhances iPSC generation from both mouse and human somatic cells. Vc acts at least in part by alleviating cell senescence, a recently identified roadblock for reprogramming
