Manipulating DNA Biology 11: Section 6.4
2
Learning Goals Students will recall bacterial conjugation Students will understand the process of recombining DNA
3
Bacterial Conjugation Form of sexual reproduction 2 or more cells fuse and plasmids are exchanged Scientists have taken advantage of this and use this to insert DNA from one organism into the DNA of another organism (genetic engineering) This led to the development of recombinant DNA technology
4
Recombinant DNA DNA that is formed when DNA fragments from 2 or more different organisms are spliced together in a laboratory How insulin for human use can be produced The human gene for insulin production was inserted into bacterial plasmid DNA and the bacterial cell expressed the gene, and started producing insulin
5
Steps for Recombining DNA 1.A restriction enzyme is used to “cut” the DNA from one organism at a specific site. This leaves unpaired nitrogen bases at the ends, called “sticky ends”. 2.The DNA fragment from the organism is put into the plasmid DNA. Bonding occurs between nitrogenous bases of the sticky ends resulting in recombinant DNA called chimeric DNA
