Biochemical tests part 2 Gr C: Explain how to do an emulsion test and a Vitamin C test. Gr B: Create a method for each test. Gr A: Analyse different foods for their contents. Entrance Activity Mini Methods Write down mini methods for the bendicts test, Biuret reagent test, and the iodine test. Key Terms Emulsion Vitamin C DCPIP Ethanol

Solution tests Biuret Reagent (Used to test for Protein) Iodine Solution (Used to test for Starch) Benedict’s reagent (Used to test for Glucose) Positive Biuret Reagent Test Start Colour:Final Colour: Positive Iodine Test Start Colour:Final Colour: Positive Benedict’s Reagent Test Start Colour:Final Colour: Clear Blue Straw Yellow Purple Orange Blue/Black Experiment

Emulsion Test 1.Measure 2 grams of unknown solution and place into a test tube. 2.Add 2ml of ethanol to the unknown solution and gently shake for 1 to 2 minutes. 3.Using a measuring cylinder, fill a separate test tube up with an equal amount of distilled water. 4.Gently pour the ethanol and unknown substance solution into the distilled water test tube. 5.If lipids are present then they should form a milky-white emulsion layer on the top.

Vitamin C Test 1.Using a measuring cylinder, measure out 8ml of DCPIP(Dichlorophenolindophenol) solution. 2.The DCPIP should be a blue colour, using your pipette take a sample from the unknown solution and begin adding drops into the DCPIP. If vitamin C is present the DCPIP will turn clear. 3.The more drops of the unknown solution you add the lower the concentration of vitamin C is in the unknown solution.

Vitamin C Test Emulsion Test I know that fats and/or lipis are present because… I know vitamin C is present because… I know how concentrated the vitamin C is in the unknown solution because… Learning Activity Complete the following sentences in your book to prove you have an understanding of each test. When I pour the unknown solution, mixed with some ethanol, onto distilled water, it forms a milky- white layer on the surface of the water. When Vitamin C is added to DCPIP it turns from blue to clear. Few drops- Strong vitamin C solution. Many drops- Weak vitamin C solution.

Learning Activity Write down a method for each of the two tests you just completed. It needs to be a very simple method, which can easily be followed, correctly, by anyone. Try and include information displayed in the success criteria. Success Criteria Simple, numbered, step by step method. Method include amounts and measurements. Equipment is described and explained why it is being used. Include additional information regarding health and safety.

Learning Activity What are the solutions made of? In front of you are 5 different solutions, each solution has a mixture of different biochemicals placed within them. Your task is to find out which biochemicals are in each one. SolutionBenedicts test Biuret reagent Iodine testContents A B C Place a tick into the box if the chemical is present and place a cross if it isnt. There are a total of 25 tests to do, so you’ll need to work as a team and assign each person a job to do.

Quick Check 1)Name the 5 biochemicals tests. 2)What is hydrolysis and condensation? 3)Define what an organic compound is. 4)Which test should be done to identify protein? What is the colour change? 5)Name the 4 groups of organic compounds. 6)How are fats and lipids drifferent?
