BLOOD ALCOHOL Principle ADH
Ethanol + NAD Acetaldehyde+NADH+H+ Alcohol dehydrogenase (ADH) catalyzes the oxidation of ethanol to acetaldehyde with the concomitant reduction of NAD to NADH2. The change in absorbance at 340nm is directly proportional to the alcohol concentration in the sample.

SPECIMEN COLLECTION AND STORAGE
The site of venipuncture should be disinfected only with aqueous disinfectants such as zephiran or merthiolate. Alcohol or other volatile disinfectants must not be used. Whole blood may be collected using citrate, heparin or oxalate as the anticoagulant.3 However, fluoride used at 5mg/dl of blood is the best preservative. Samples may be stored at 2-80C for several days and may not show appreciable loss of alcohol, when well stoppered.4 Serum samples may be assayed without deproteinization. Serum Cholinestrese

Collection of Samples Urine A urine specimen should be obtained, before the administration of any drugs. Usually a 50ml sample without added preservatives is sufficient for a comprehensive series of tests. Stomach Contents It is very important to collect and send the first sample of washing, atleast 50ml to carry out a wide variety of tests, without added preservatives. Blood Samples which should be taken on admission of the patient are 10ml of heparinised blood
