Epithelial tissues of normal urinary bladder (control, C) and transitional carcinomas with various tumor grades (G) were collected, processed in protease inhibitor containing lysis buffer, and vigorously equal amounts of protein were subjected to SDS-PAGE to perform Western blot analysis of the PKC isoforms as described under “Materials and methods”. (A) Representative Western blot data of several determinations yielding similar results. (B) The amounts of the expressed PKC isoenzymes were quantitated by densitometry (optical density; OD), and values obtained with the tumor samples were expressed as the percentage of the control samples (normalized OD), regarded as 100% (solid line). Points represent mean±SEM of several independent experiments for each antigen. The ∗ represents significant (p<0.05) differences of OD values of any tumor sample compared to control. The ∗∗ represents significant (p<0.05) differences between the OD values of the G2 or G3 tumors compared to that of the G1 samples.
