1 HIGH RESOLUTION IMAGING FACILITY (HRIF)
TRANSMISSION ELECTRON MICROSCOPY (TEM) Kent Keyser, Ph.D. Director (Office) Melissa Chimento, TEM Core Manager (Office) The Transmission Electron Microscopy (TEM) HRIF Core provides access to conventional transmission electron microscopy and associated sample processing services. The facility is staffed by experienced technical specialists who are able to offer assistance at each stage of an imaging experiment. To schedule time in the lab or to schedule a training session with an HRIF specialist, visit Recent Publications with Tecnai T12 TEM Core Equipment FEI TECNAI SPIRIT T12 High Voltage range kV Embedded AMT XR60B CCD Camera Suitable for imaging thin sections (~70nm) of tissue and cells, negatively stained viruses, bacteria and materials science samples Dr. Sontheimer and his colleagues used “a clinically relevant mouse model of glioma and find that glioma cells, as they populate the perivascular space of preexisting vessels, displace astrocytic end feet from endothelial or vascular smooth muscle cells. This causes a focal breach in the Blood Brain Barrier(BBB). Furthermore, astrocyte-mediated gliovascular coupling is lost, and glioma cells seize control over the regulation of vascular tone through Ca(2+)-dependent release of K(+). These findings have important clinical implications regarding blood flow in the tumor-associated brain and the ability to locally deliver chemotherapeutic drugs in disease”. “Disruption of astrocyte-vascular coupling and the blood-brain barrier by invading glioma cells”. Leica UC6 & UC7 Ultramicrotomes with Cryo Kit Leica ultramicrotomes provide easy preparation of semi- , ultrathin, and cryo sections, as well as perfect, smooth surfaces of biological and industrial samples for TEM, SEM, AFM and LM examination. Leica EM AFS2 (Freeze Substitution Apparatus) The Leica EM AFS2 performs freeze substitution and progressive lowering of temperature (PLT) techniques and allows low temperature embedding and polymerization of resins. Ted Pella BioWave Microwave Tissue Processor Microwave stimulated rapid processing of tissues for histopathology and electron microscopy processing with the conventional Heme oxygenase-1-mediated autophagy protects against pulmonary endothelial cell death and development of emphysema in cadmium-treated mice Pulmonary exposure to cadmium, a major component of cigarette smoke, has a dramatic impact on lung function and the development of emphysema. Cigarette smoke exposure induces heme oxygenase-1 (HO-1), a cytoprotective enzyme. In this study, we employed a truncated mouse model of emphysema by intratracheal instillation of cadmium (CdCl2) solution (0.025% per 1 mg/kg body wt) in HO-1(+/+), HO-1(-/-), and overexpressing humanized HO-1 bacterial artificial chromosome (hHO-1BAC) mice. Pretreatment of HO-1(+/+) PECs with rapamycin induced autophagy and resulted in reduced cell death upon cadmium treatment. Induction of autophagy following CdCl2 treatment was found to be protective from apoptotic cell death. HO-1 induced protective autophagy in PECs and mitigated cadmium-induced emphysema.
