The Microscope
2
History… 1590 Hans and Zacharias Jensen Dutch eyeglass makers
Designed first microscope 1600’s Leeuwenhoek Made lenses as a hobby Made over 250 microscopes Made microscope that could magnify 300 times First to see living organisms “animalcules”
3
Tools to view specimen-
1. Hands lens (magnifying glass)- magnifies 3-5x
4
Tools to view specimens-
2. Stereoscope (dissecting scope)- magnifies 10-60x; shines light onto the specimen; solid objects
5
Tools to view specimens-
3. Light microscope (compound)- magnifies up to 2,000x (ours 400x), light passes through the specimen and then through 2 or more lenses
6
Tools to view specimens-
Scanning electron microscope (SEM)- 50,000x; surface of specimen coated with metal; electrons bounce off and 3D image is recorded
7
Tools to view specimens-
5. Transmission electron microscope (TEM)- 2,000,000x; specimen has to be sliced thin; electrons pass through it; 2D image is recorded
8
Total Magnification: Light microscope- eyepiece x objective lens = total mag Example: 10 (always) x 40 = 400x
