Tissue transglutaminase autoantibody enzyme-linked immunosorbent assay in detecting celiac disease  Satu Sulkanen, Tuula Halttunen, Kaija Laurila, Kaija-Leena Kolho, Ilma R. Korponay-Szabó, Annikki Sarnesto, Erkki Savilahti, Pekka Collin, Markku Mäki  Gastroenterology  Volume 115, Issue 6, Pages (December 1998) DOI: /S (98) Copyright © 1998 American Gastroenterological Association Terms and Conditions

2 Fig. 1 Western blot analysis of tissue transglutaminase with monoclonal antitransglutaminase antibody (lane 1), control sera (lanes 2–4), and celiac disease sera (lanes 5–8) under (A) nonreducing and (B) reducing conditions. Positions of molecular mass standard (kilodaltons) are indicated. Gastroenterology  , DOI: ( /S (98) ) Copyright © 1998 American Gastroenterological Association Terms and Conditions

3 Fig. 2 Serum IgA class tissue transglutaminase antibody (IgA-tTG-ab) titers determined by ELISA. The results are presented as AU values. The filled diamonds given an AU >100 represent individual values from 105 to 663 AU. The following patient groups were studied: I, newly diagnosed untreated celiac patients (n = 136); II, patients with celiac disease during gluten-free diet (n = 38); III, celiac patients during gluten challenge (n = 18); IV, again after gluten withdrawal (n = 11); and V, disease controls with normal small bowel mucosa (n = 207). The points in the figure connected with the line represent the values of the same patients in the different groups. An arbitrary cutoff level for positivity (dotted line) was drawn at an AU of 10. The box in group V represents 194 individuals with negative results. Gastroenterology  , DOI: ( /S (98) ) Copyright © 1998 American Gastroenterological Association Terms and Conditions

4 Fig. 3 Correlation between serum IgA class tissue transglutaminase (IgA-tTG-ab) and endomysial (IgA-EmA) autoantibodies. ♦, Patients with untreated celiac disease; 2, control patients with normal jejunal mucosa. The box represents 194 disease control patients with negative results on both tests. The filled diamonds given an AU >100 represent individual values from 105 to 663 AU. 5 Fig. 4 Human umbilical cord was double immunostained with (A) untreated celiac disease patient sera (green) and (B) monoclonal antitransglutaminase antibody (red). (C) These two staining images were merged, and the colocation of the antigens is represented by yellow. (D) Double immunostaining with nonceliac serum with anti–smooth muscle antibodies (green) and with monoclonal antitransglutaminase antibody (red).
