3. Human HO-1: general introduction
Heme oxygenase (HMOX1) cleaves the heme ring at the alpha methene bridge to form biliverdin. Biliverdin is subsequently converted to bilirubin by biliverdin reductase.
Stressed mammalian cells up-regulate heme oxygenase 1 (HO-1), which catabolizes heme to biliverdin, carbon monoxide, and free iron. HO-1 plays a crucial role in the inflammatory process during wound healing.
Structure of human HO-1
TNFalpha accelerates inflammatory responses by down-regulating HO-1 expression in human monocytes. TNF antagonists may block this TNF-dependent suppression of HO-1 expression, resulting in an amelioration of inflammation
Defects in HMOX1 are the cause of heme oxygenase 1 deficiency (HMOX1D). A disease characterized by impaired stress hematopoiesis, resulting in marked erythrocyte fragmentation and intravascular hemolysis, coagulation abnormalities, endothelial damage, and iron deposition in renal and hepatic tissues.
