Measurement of PTP opening into PTP-containing proteoliposomes. a. Dose-response of calcium (Ca2+). 10µL of plain or PTP-containing liposomes were treated by various doses of Ca2+ for 30 min at 25°C and the release of 4-MUP from the liposomes has been determined by fluorimetry as described in material and methods. Results are expressed as percent of the Ca2+ producing the maximal 4-MUP release. b. Inhibition of PTP opening by ATP. 10µL of PTP-containing liposomes were pretreated with ATP for 15 min at 25°C before Ca2+. c. Tat peptide effect on PTP. 10µL of plain or PTP-containing liposomes are treated by various doses of Tat in same conditions and results are expressed as percent of 2.1 mM Ca2+. The HIV-1 derived Vpr52-96 protein was used as a positive control in the same experiment. The results are representative of three independent experiments. Supplementary Figure S1, Lecoeur et al.
