12 What’s going on in there? Smaller pieces travel faster

13 CRIME SCENE – Ms. Yeung’s desk.

14 CRIME SCENE – Ms. Yeung’s desk.
WHO CHANGED ALL THE GRADES IN MS. YEUNG’S GRADEBOOK INTO F’S??

15 EVIDENCE #1 - Foot prints
#2 #3 #1

16 Suspect #1 K. Minor last seen making a sandwich at Ms. Yeung’s desk
Motive – new teacher, may have been an accident she’s unwilling to admit to

19 EVIDENCE #2 – Strand of hair

20 How do you identify a suspect?
Restriction Enzymes – enzymes that cut up DNA segments at a particular sequence of bases ie. EcoR1 (originated from E. coli) G A A T T C C T T A A G

21 How do you identify a suspect?
Each suspect’s DNA will be cut into a combination of long and short strands that will be unique to that suspect T T G A A T T C G G C G G A A T T C A T A G G C T G G G C C A A C T T A A G C C G C C T T A A G T A T C C G A C C C G G T T G G C C G A A T T C G G C G G C A T T C A T A G G C T G A A C C C G G T T A A G C C G C C G T A A G T A T C C G A C

22 How do you identify a suspect?
We’ll use gel electrophoresis to separate out the different sized DNA segments Each suspect will have a unique pattern on the gel that we will compare to the DNA from the crime scene (CS)

23 How do you identify a suspect?
U = Uncut (did not add restriction enzyme) How many bands should we expect to see on our gel for the “U” tube? Only 1 because the DNA strand did not get cut Should the band travel close to the well or far from the well? Close, because it’s one long strand
