B, iTRAQ experimental design. Spheroids are grown, in biological duplicate (25 spheroids for each sample), subject to either no glucose or no FBS, homogenized, and harvested for protein. Subsequent proteins are digested into tryptic peptides. The primary amines of the peptides are labeled with 6 unique isobaric tags with specific reporter ion m/z values. Labeled digests are mixed into one sample. The combined sample is then analyzed using LC–MS/MS to quantitate and identify proteins in different biological samples.
