Selection for fit types in a variable population
Selection pressure: antibiotic Mutants survive Are NalR mutants less fit in an antibiotic-free environment? Tradeoff?
3
Nucleotide sequence of candidate genes Mutations?
Select for NalR mutants (different Nal-Conc) correlation between selective pressure and number of mutants? Nucleotide sequence of candidate genes Mutations? Recovery of fitness during many generations when selection removed? Duration? Nal class of quinolones;inhibiy DNA gyrase (gyrA and gyrB) and Topoisomerase IV (parE and parC) affect DNA supercoiling

Elsa: we would expect to see mutations in this gene, we only see one, however we only have part of the sequence for this gene. Alternately other mutations could confer resistance, or it could have been lost during isolation/culture.

Suggestions: Maintain selective pressure for NalR Mutant up to “cage fight” ? Plate replicates and/or various timepoints
