Purpose of DNA Replication/Processes
The purpose of DNA replication is mostly to pass down genetic material. It also to makes a copy of its parent DNA. DNA replication occurs during the S- phase portion of the interphase in mitosis. For DNA to start the double helix has to unwind. The DNA Helicase then comes in and and splits the nucleotides and breaks the hydrogen bonds between them. Next, RNA Primase comes in and puts down a RNA primer which tells the DNA Polymerase III where to go to add the missing nucleotides. Then DNA polymerase I comes in and makes the primer into DNA. The final stage in strand I is that DNA Ligase stitches the two nucleotides together. In strand II, basically the same things happen. RNA Primase comes in and makes several RNA primers and then DNA polymerase III comes in and makes the other nucleotides. Then, DNA polymerase I makes the primers into DNA and then the ligase comes in and stitches them together.

Mutations A mutation, which may arise during replication and/or recombination, is a permanent change in the nucleotide sequence of DNA. Damaged DNA can be mutated either by substitution, deletion or insertion of base pairs. Mutations, for the most part, are harmless except when they lead to cell death or tumor formation. Some mutations could include base substitutions, deletions, and insertions. If mutations occur, things that could happen in the DNA are errors in DNA replication, errors in DNA recombination, chemical damage to DNA, and radiation.

Extra Information Telomeres are a compound structure at the end of a chromosome. Telomeres are crucial to the life of the cell. They keep the ends of the various chromosomes in the cell from accidentally becoming attached to each other. Okazaki fragments are short, newly synthesized DNA fragments that are formed on the lagging template strand during DNA replication. DNA ligase is an enzyme that stitches the nucleotides together. Telomerase is an enzyme that adds nucleotides to telomeres, especially in cancer cells. Cancer cells may be immortal and in culture produce telomerase. Cancer cells show no contact inhibition. Once the surface of the dish is covered, the cells continue to divide, piling up into mounds. The use of stem cells as a treatment for cancer or other illness is in stem cell transplantation. Cloning is when you replicate (a fragment of DNA placed in an organism) so that there is enough to analyze or use in protein production. Aging is the process of getting old. If lack of telomerase hastens replicative senescence, it may also hasten the aging of the tissues that depend on newly-formed cells for continued health — a tradeoff that may not be worth making.
