NBC Seminar Toward the Cell Factory : Carbohydrate Biorefinery of Thermus caldophilus GK24 Speaker: Dae-Sil Lee, Ph.D School of Biological Sciences, University of Ulsan Thermus caldophilus GK24 (TCA) is an extremothermophile growing at 70 Co, providing industrially important enzymes. TCA genome sequence has been determined, and annotated by home-made system (Biopot). Then the genes responsible for carbohydrate-related enzymes were identified, expressed in E. coli and characterized. In this lecture, ‘Thermus-derived Cell Factory’ consisted of  130 thermostable enzymes will be introduced, particularly focusing on in vitro operation of glycolysis and pentose-phosphate pathway to give the synthetic building block as well as the glycolytic products. The application of this synthetic network could be extended to synthesis of sedoheptulose-7-phosphate from starch containing a-glucan phosphorylase, phosphoglucomutase, glucose-6-phosphate isomerase, transaldolase and transketolase. Overall, 'Thermus Cell Factory' with thermostable carbohydrate enzymes turned out to be an excellent tool to synthesize various C3-C9 phosphosugars and industrial metabolites, as well as to search novel metabolic pathways and the molecules governing the regulation of metabolisms.
