Temperature…. Depend on body temperature of animal from which cell is obtain. Depend on part of body (temperature of skin and testis lower than rest of body). Birds and avian-38.5*c. Human and warm blooded animals-37*c. Cold-blooded animals-15*c-26*c. Cell can`t tolerate high temperature than normal.

OSMOLALITY…. Salt concentration of cell. Human plasma-290miliosmo/kg
Mice-310miliosmo/kg miliosmo/kg is optimum . Measured by depression of freezing point or elevation of vapour pressur of medium. Hepes ,acids and bases are used for regulation of osmolality.

FOAMING…. Characteristic of suspension. Denatration of proteins.
Interfare with the exchange of gas phase. To prevent foaming add antifoaming agent ex: pluronic F68,carboxy methyl cellulose CMC).

VISCOSITY….. Prevent cell damage(cell agitation or after trypsinization). Increase viscosity by adding carboxymethlycellulose or polyvinylpyrrolidone(PVP) AND SERUM.
