Background Plum curculio is a pest of stone fruit in eastern North America There are two strains: northern univoltine and southern multivoltine The southern multivoltine strain is a source of concern because second generation larvae may be present in the fruit at harvest

Observations (1) mtDNA can distinguish Byron samples from other samples (2) There are three Wolbachia strains (w1, w2, w3) along the Eastern U.S. (3) Only one strain, w3, has been identified to date in GA (4) RFLP can be used to determine which Wolbachia strain is causing infection

Conclusions Total sequencing expense is more than RFLP expense.
Sequencing cost is $8 - $12 more per sample than RFLP. PCR-generated RFLP fingerprints provide promise as a low cost alternative to sequencing, but need further investigation.
