1 Cravings? Fix your leptin!

2 Functions: regulating appetite, food intake and metabolism.
Adipose Derived Hormone Functions: regulating appetite, food intake and metabolism. Also regulates puberty onset and pregnancy. Secreted by adipose tissues Counteracts the effects of: neuropeptide Y in the gut (feeding stimulant) cannabinoid neurotransmitter, adandamide (stimulates appetite) Promotes synthesis of: α-melanocyte-stimulating hormone (appetite suppressant) Adipose tissue absorbs glucose, & lipids 2. Releases leptin 3. Leptin binds to neurons in hypothalamus 4. α-melanocyte-stimulating hormone synthesizes Result: sense of satiation Sources: (“Cravings?” “Leptin,” “The Stomach-”)

3 Homeostatic control of adipose tissue mass
Fat mass fails -> plasma leptin concentration falls -> stimulating appetite Fat mass increases -> leptin levels increase -> suppressing appetite Leptin is the afferent signal in a negative feedback loop that maintains homeostatic control of adipose mass. It circulates in the blood and acts on the brain to regulate food intake. When fat mass falls, plasma leptin concentrations fall too, stimulating appetite and suppressing energy expenditure until fat mass is restored. When fat mass increases, leptin levels increase, suppressing appetite until weight is lost. This system maintains homeostatic control of adipose tissue mass. Leptin acts on a receptor localized in the hypothalamus, and elsewhere in brain, to regulate energy balance and other (Source: Nature Medicine)

4 Leptin resistance Analogy Air-conditioning Too hot and heating stops
High levels of leptin = “Brain is starved, the body is obese” Heart disease, obesity, diabetes Source: (Leptin Hormones) Analogy Air-conditioning Too hot and heating stops Too cold and heating begins
