Enzyme-linked immunosorbent assay (ELISA) for desmoglein 3. Anti-Dsg3 antibodies (αDsg3) from pemphigus serum binds Dsg3 on the ELISA plate; irrelevant antibodies, that do not bind, are washed off. The plate is then incubated with horseradish peroxidase (HRP) conjugated antihuman IgG, which binds the anti-Dsg3 IgG that is on the plate. HRP is an enzyme that turns a clear substrate blue and the amount of color, read on spectrophotometer, correlates with the amount of pemphigus (i.e., anti-Dsg3) antibody in the patient's serum.
