1 From: Controlled Delivery of 5-Chlorouracil Using Poly(Ortho Esters) in Filtering Surgery for Glaucoma Invest. Ophthalmol. Vis. Sci ;49(7): doi: /iovs Figure Legend: Analysis of annexin-V binding. Rabbit Tenon fibroblasts were exposed to 5 μg/mL concentration (A, B, E, F) or 100 μg/mL concentration (C, D, G, H) of 5-CU or 5-FU. Annexin-V binding was tested after 24 hours of exposure. (A) Annexin-V with DAPI staining of cells exposed to 5 μg/mL 5-CU. (B) Annexin V binding. White arrows indicate cells with diffuse binding of Annexin V on the cell surface suggesting phosphatidylserine exposure. (C) Annexin V with DAPI staining of cells exposed to 100 μg/mL 5-CU. (D) Annexin V staining of the same cells. White arrow indicates a cell with condensed nucleus and diffuse surface binding of annexin V. (E) Annexin V with DAPI staining of cells exposed to 5 μg/mL 5-FU. Higher magnification of the nucleus shows chromatin alteration. (F) Annexin staining of the same cells showing clear cytoplasmic accumulation of annexin V inside the cytoplasm, suggesting enhanced cell permeability. (G) Annexin V with DAPI staining of cells exposed to higher dose of 5-FU. (H) Annexin staining of the same cells showing clear cytoplasmic accumulation of annexin V within the cytoplasm, suggesting enhanced cell permeability (white arrows). (I, J) No annexin V binding in control cells. Scale bar, 20 μm. Date of download: 10/23/2017 The Association for Research in Vision and Ophthalmology Copyright © All rights reserved.
