Anton van Leeuwenhoek ( ) Discovered Bacteria Father of the Modern Microscope
3
Characteristics of Bacteria
Simplest Living Organisms Single Celled: Prokaryote (no true Nucleus) Reproduce by Fission (dividing into 2) Found Everywhere Nutrition Chemotrophs - eat by absorbing chemicals Phototrophs - make food by photosynthesis
4
Bacteria Structure
5
CLASSIFICATION 1. By shape a) Cocci (Spherical)
b) Bacillus (Rod-shaped) c) Spirillum (Spiral Cell)
6
Cocci Spherical

CLASSIFICATION Arrangements Staphylo- = Clusters Strepto- = Chains
Prefixes Staphylo- = Clusters Strepto- = Chains Diplo- = Twos
12
Staphylococcus aureus
13
Streptococcus pyogenes
14
Diplococcus
15
Anthrax (Bacillus anthracis)
16
CLASSIFICATION 2. By Cell Structure Simple Staining - tells shape
We use 1 stain. Grams Staining - to see cell wall structure
17
Gram-negative rods
18
Aerobic, Gram-positive rods
19
Aerobic, Gram-positive cocci
20
Anaerobic, Gram-positive rods

Growth Requirements Food - Water - Warmth - (Store in Incubator)
agar contains all Nutrients Water - agar, powder & water mixture Warmth - (Store in Incubator) optimum temp. for most rapid growth is: degrees C degrees F
23
Culturing To provide all the requirements for growth
Usually requires hours for growth Each dot (.) that appears is a COLONY, growth started from 1 or more bacteria cells
24
Visible colonies of bacteria in a petri plate.

The Good With The Bad BAD = GOOD =
Bacteria can cause disease and infections. Ex. Pneumonia, scarlet fever, TB, Anthrax GOOD = Decompose materials “Nature’s Recycler” Make plastics, pesticides, oil eating bacteria
28
I hope you enjoyed your introduction into the wonderful world of….
BACTERIA
29
Place a slide with a bacterial smear on a staining rack.
STAIN the slide with crystal violet for 1-2 min. Pour off the stain. Note: fingers stain Gram-positive - use forceps! Flood slide with Gram's iodine for 1-2 min. Pour off the iodine. Decolourize by washing the slide briefly with acetone (2-3 seconds). Wash slide thoroughly with water to remove the acetone - do not delay with this step. Flood slide with safranin counterstain for 2 min. Wash with water. Blot excess water and dry in hand over bunsen flame.
