1 Optimization of factors for production of  extracellular hemicellulases and pectinase by a chromium resistant  strain of Trichoderma pseudokoningii. 2 In brief about them… Hemicellulase is a type of enzyme that degrades hemicellulose. It includes Cellulases and Xylanase. Pectinase are group of enzymes that include polygalacturonase, pectin lyases and pectin methyl esterase and catalyses the degradation of pectic substances

3 Applications Hemicellulase is used commercially in baking products and in various other food preparation technologies. It is taken by some as a supplement to aid in digestion Pectinase have been used in several conventional industrial processes, such as textile, plant fiber processing, tea, coffee, oil extraction, treatment of industrial wastewater, containing pectinacious material, etc. They have also been reported to work on purification of viruses and in making of paper. They are yet to be commercialized.

6 Aims & Objectives Whether a chromium resistant strain is capable of producing all three important hemicellulases(β –glucosidase, endoglucanase and endoxylanase) and pectinase (polygalacturonase). Optimization of factors for achieving maximum production of these enzymes.

7 Effect of Various Substrates
Pectinases and hemicellulases (β –glucosidase, endoglucanase and endoxylanase) shows the maximum production.

(β –glucosidase, endoglucanase and endoxylanase)

9 Effect of Substrate Concentration
β –Glucosidase and Endoglucanase showed its optimum at 1.0% (w/v) using pure cellobiose and CMC; Endoxylanase showed its optimum at 0.5%.

10 Effect of Temperature Most favorable production temperature for hemicellulase (β –Glucosidase, endoglucanase, endoxylanase) production was found to be 27°C

11 Effect of pH Maximum enzyme activity was obtained when the initial pH of the production media supplemented with β –Glucosidase and endoglucanase were adjusted to 5.0 and endoxylanase to 6.0

12 Effect of Cultivation time
All hemicellulase enzymes (β –Glucosidase, endoxylanase and endoglucanase) showed maximum cultivation period after 24 hours (1 day)

13 Effect of various nitrogen sources
Of the various nitrogen sources used, maximum β –Glucosidase activity was observed when gelatine was used, although in case of endoglucanase and endoxylanase their enzyme activity was maximum on using peptone and potassium nitrate respectively. Urea and ammonium sulphate reduced the hemicellulase production in almost all types of substrates.

Rapid producer of enzymes Different enzymes could be produced Pectinase produced is : With high pH and temp stability So far the literature study is concerned, this is the first report of hemicellulase and pectinase production from a chromium tolerant strain of Trichoderma sp.
