1. In 1st image I got desired band at 3 cycle (high efficieny, 30 sec on and 45 sec off) but in next image I got at 4 cycle (high efficieny, 30 sec on and 45 sec off) or at 3 cycle (high efficieny, 60 sec on and 45 sec off).
And so my results are not consistent.

2. 12_02_14 With decreased contrast
Sonication cycle bp
Formaldehyde 2%
Agarose gel 1.5%
From ladder side 1st 3 lanes are WT( high efficiency) and next three are p100-/-(low efficiency sonication cycles)
Reverse crosslink for 5 hrs
RNase treated

3. Sonication 100bp
Cycle (………………30 on ) (45 on) (60 on) (……….30 on…….) (15 on) (45 on) (60 on)

4. 1st six lanes are WT and next six lanes are P100-/- 3T3 cells, Formaldehyde 2%
Agarose gel 1.5%, Reverse crosslink for 5 hrs at 65oc, RNase treated, all at high efficiency,
All other conditions were same as I mentioned before.
