1
Screening of Anti-inflammatory Drugs
Lab # 4
2
Outline Describe the inflammatory response
Anti-inflammatory drugs; MOA, Uses, SE Explain methods for screening of Anti-inflammatory drugs Discuss the lab work Gamra Hamad
3
Inflammation Response of immune system to foreign organism or antigenic substance Characterized by edema, erythema, swelling, and pain or tenderness Gamra Hamad
4
Inflammatory Mediators
Phospholipids Gamra Hamad
5
Phases of inflammation
Acute: vasodilation & increase capillary permeability Delayed: infiltration of leukocytes and phagocyte cells Chronic proliferative: tissue degeneration and fibrosis Gamra Hamad
6
Anti-inflammatory Drugs
Steroidal: ex. Corticosteroids (inh. gene transcription ) Non-steroidal Anti-inflammatory Drugs (NSAID) (inh. COX) Examples: ASA, indomethacin, ibuprofen Selective COX-2 inhibitors Gamra Hamad
7
NSAID Uses: rheumatoid arthritis
SE: GI adverse effects, renal insufficiency, bronchospasm Gamra Hamad
8
Screening methods For Anti-inflammatory Drugs
Granuloma Pouch edema methods Croton oil Control vs. treated Calculate % inhibition Adjuvant-induced arthritis ID Tubercle bacilli Calculate % change in edema & body wt. over 14 days Gamra Hamad
9
Screening methods For Anti-inflammatory Drugs
Rat-paw edema Animal 6 rats (3 control, 3 treated) Drugs: carragenin (2%, 0.1 ml SC.), indomethacin 2 mg/kg IP Gamra Hamad
10
Cont. Control group Treated group Rt. Lt. 1 2.9 1.5 1.9 1.3 2 3.1 1.7
1.4 3 1.6 Mean A B C D Gamra Hamad
11
Calculation % of edema produced by carragenin alone
= A – B X = X % B % of edema produced by carragenin after ttt with indomethacin = C – D X = Y % D % reduction of edema by indomethcin = X –Y= % Gamra Hamad
