Blockade of NADPH Oxidase Restores Vasoreparative Function in Diabetic CD34+ Cells
Invest. Ophthalmol. Vis. Sci ;52(8): doi: /iovs Figure Legend: NADPH oxidase activation and superoxide levels are higher in diabetic CD34+ cells. (A) Expression of Nox2 mRNA relative to β-actin was higher in diabetic CD34+ cells than in nondiabetic cells (*P < 0.01, n = 10). (B) NADPH oxidase activity in the lysates of diabetic CD34+ cells, expressed as AFU per milligram protein, was higher than in nondiabetic cells (*P < 0.01, n = 5). (C) Intracellular levels of superoxide determined by quantitative fluorescence imaging using DHE. Shown are images of red fluorescence emitted by the oxidized product of DHE, 2-hydroxyethidium (HEt) after DNA intercalation. Images were obtained with a Fluor 40×W objective. (left) Color scale for the fluorescence intensity. Scale bar, 20 μm; applicable for all images. (D) Box plots of HEt fluorescence observed in diabetic and nondiabetic CD34+ cells. HEt fluorescence was higher in diabetic CD34+ cells than in nondiabetic cells (*P < , n = 4, Mann-Whitney test).
