(A) Expression of Nox2 mRNA relative to β-actin was higher in diabetic CD34+ cells than in nondiabetic cells (*P < 0.01, n = 10). (B) NADPH oxidase activity in the lysates of diabetic CD34+ cells, expressed as AFU per milligram protein, was higher than in nondiabetic cells (*P < 0.01, n = 5). (C) Intracellular levels of superoxide determined by quantitative fluorescence imaging using DHE. Shown are images of red fluorescence emitted by the oxidized product of DHE, 2-hydroxyethidium (HEt) after DNA intercalation. Images were obtained with a Fluor 40×W objective.
